Khaleel Ahmed was the Sunrisers Bowler’s pick 3-21.
Bairstow is now the third highest runs scorer after Shikhar Dhawan of Delhi Capitals and Glenn Maxwell of Royal Challengers Bangalore in the competition this season with 173 runs over four innings.
He hit three fours and three sixs in his 56-ball innings and it took 48 balls to hit his half-century – the longest he has ever taken in a Twenty20 match.
It was his seventh half century in IPL cricket and the highest individual chase score in Chennai that season.
Sunrisers are the third team in eight games to win the Chennai Pursuit this season.
The win brings Sunrisers – who are trained by English World Cup winner Trevor Bayliss – to fifth place in the eight-team table, while Punjab is now in last place.
